Hypothalamische Entzündung und metabolisches Syndrom

Zusammenfassung

Übergewicht und Adipositas spielen eine zentrale Rolle in der Entstehung des metabolischen Syndroms. Seit einigen Jahren wird intensiv untersucht, wie Störungen des Zentralnervensystems das Essverhalten, die Entwicklung von Übergewicht und Adipositas sowie Störungen der Glukosetoleranz beeinflussen können. In Tiermodellen, aber auch beim Menschen konnte gezeigt werden, dass eine energetische Überversorgung mit fettreicher Nahrung zu weitreichenden strukturellen und funktionellen Veränderungen in hypothalamischen Neuronen führt. Beispielsweise konnte belegt werden, dass Apoptose von hypothalamischen Neuronen und Gliazellen sowie die sich anschließenden lokalen Entzündungsreaktionen die Nahrungsaufnahme und den Stoffwechsel beeinflussen. Somit begünstigen sie Übergewicht und die Entstehung des metabolischen Syndroms. Dieser Beitrag fasst aktuelle Studien zu dieser Thematik zusammen, stellt einige der zugrunde liegenden molekularen Mechanismen vor und zeigt auf, wie dieses Wissen möglicherweise zu neuen Therapieoptionen für die Behandlung von Adipositas und metabolischem Syndrom führen kann.
