Zusammenfassung

Die Multiphotonentomographie (MPT) respektive Multiphotonenmikroskopie (MPM) hat in den letzten Jahren zunehmende Bedeutung als nichtinvasive Untersuchungsmethode in der Dermatologie erfahren. Die Bildgebung mithilfe der MPT basiert auf der spezifischen Anregung natürlich in der Haut vorkommender, biogener Fluorophore. Daneben kann der Effekt einer Induktion der Frequenzverdopplung bestimmter Moleküle für die Bildgebung genutzt werden. Damit ist es möglich, auf eine zusätzliche Fluoreszenzmarkierung von Geweben zu verzichten – mit entscheidenden Vorteilen für die Beurteilung von Hautveränderungen in vivo. Das Verfahren wird sowohl in der klinischen Diagnostik als auch in der biomedizinischen Forschung eingesetzt. Durch die MPT wird eine „optische Biopsie“ bis in Tiefen von 200 µm ermöglicht, die zelluläre und extrazelluläre Strukturen mit subzellulärer Auflösung darstellt. In Kombination mit der Fluoreszenzlebensdauer angeregter Fluorophore ergeben sich ergänzende Informationen über das Mikroenvironment, energetische Zustände und den Zellmetabolismus. In diesem Review werden die aktuellen Entwicklungen der In-vivo-Diagnostik mithilfe der MPT bei der Beurteilung physiologischer und pathologischer Veränderungen der Haut vorgestellt.
