Is late-onset disease or the lower rate of M694V mutations associated with the mild disease phenotype?

We read with interest the article by Ozdel et al. [
1] indicating that late-onset disease is associated with a mild phenotype in children with familial Mediterranean fever. We have a comment on this study, which would facilitate interpretation of the findings and a potential and important limitation. It is well known that M694V mutation is associated with a younger onset and more severe disease phenotype [
2]. It can be clearly seen that the group with late-onset disease has a significantly lower rate of M694V mutations. Furthermore, the authors did not seem to perform a multivariate regression, which might indicate that late-onset disease is associated with a mild phenotype independent of M694V mutation status. …
