Congestive heart failure (CHF) is known as one of the covariates [14–18]. Two studies using rats indicated that right-sided heart failure (HF) with hepatic congestion (HC) can reduce theophylline clearance, rather than left-sided HF with pulmonary congestion (PC) [19,20].

This work has ranged from early investigations of the effects of environmental factors, age, diet, and other drugs on a drug's PK183 to more recent investigations that explain the genetic basis for serious adverse drug interactions for some of the most highly prescribed drugs.184 The identification and detailed understanding of the mechanistic basis of these types of interactions provides much needed information to healthcare professionals, so that safe and effective use of these medications is possible.
