Herein, we employed L-NAME (NLG-nitro-l-arginine methyl ester), which induces hypertension via inhibition of NO synthase activity [10], to construct preeclampsia model in rats. Apocynin was used to treat the preeclampsia rats at various doses, i.e. 8, 16, and 32 mg/kg/day, using normal pregnant rats as controls.

2015, Toxicology LettersCitation Excerpt :In brief, transgenic mice with a ∼10-fold cardiac-specific overexpression of the heavy metal scavenger metallothionein were described previously (Yang et al., 2006; Ye et al., 2003). Five–six month-old adult male metallothionein transgenic mice and their wild type friendly virus B (FVB) mice were subjected to drinking water containing 0.1 g/L NG-nitro-l-arginine methyl ester (l-NAME) for 14 days to induce experimental hypertension (Bernatova et al., 2000; Bourraindeloup et al., 2004; Numaguchi et al., 1995). A subset of FVB and metallothionein transgenic mice was given the autophagy inhibitor 3-MA (10 mg/kg/days, i.p.) (Guo et al., 2012) while receiving l-NAME (0.1 g/L) simultaneously for a period of 14 days to examine the role of autophagy in l-NAME and metallothionein-induced cardiac response, if any.
