Abstract

The isolated heart is deservedly one of the most popular experimental models in cardiovascular research, both in terms of cost and the quality and quantity of data it provides. However, it is a deceptively simple model, capable of throwing many problems in the path of the inexperienced or unwary perfuser. The following article discusses the advantages and disadvantages of the various types of isolated heart perfusion (Langendorff and working; blood and buffer perfused). We attempt to give an insight into the many factors which must be taken into consideration when first establishing these preparations, we identify the range of indices that can be measured and the potential pitfalls which, with a little care, can be readily avoided.

CPA toxicity screening of cryoprotective solutions in rat hearts

In clinical practice, donor hearts are transported on ice prior to transplant and discarded if cold ischemia time exceeds ∼5 h. Methods to extend these preservation times are critically needed, and ideally, this storage time would extend indefinitely, enabling improved donor-to-patient matching, organ utilization, and immune tolerance induction protocols. Previously, we demonstrated successful vitrification and rewarming of whole rat hearts without ice formation by perfusion-loading a cryoprotective agent (CPA) solution prior to vitrification. However, these hearts did not recover any beating even in controls with CPA loading/unloading alone, which points to the chemical toxicity of the cryoprotective solution (VS55 in Euro-Collins carrier solution) as the likely culprit. To address this, we compared the toxicity of another established CPA cocktail (VEG) to VS55 using ex situ rat heart perfusion. The CPA exposure time was 150 min, and the normothermic assessment time was 60 min. Using Celsior as the carrier, we observed partial recovery of function (atria-only beating) for both VS55 and VEG. Upon further analysis, we found that the VEG CPA cocktail resulted in 50 % lower LDH release than VS55 (N = 4, p = 0.017), suggesting VEG has lower toxicity than VS55. Celsior was a better carrier solution than alternatives such as UW, as CPA + Celsior-treated hearts spent less time in cardiac arrest (N = 4, p = 0.029). While we showed substantial improvement in cardiac function after exposure to vitrifiable concentrations of CPA by improving both the CPA and carrier solution formulation, further improvements will be required before we achieve healthy cryopreserved organs for transplant.
Eucalyptol potentiates antianginal effect while blunts tachycardia of nifedipine in rats via nitric oxide, antioxidant, and anti-inflammatory pathways: In vivo and in vitro study

Eucalyptol; a terpenoid oxide present in numerous plant-derived essential oils; is used in many foodstuffs, however, its excessive use can cause serious adverse effects. This study investigated effects of the lowest effective doses of eucalyptol in vasopressin-induced ischemia in rats which simulates human angina. In vivo evaluations included ECG, blood pressure, plasma nitric oxide (NO), aortic endothelial NO synthase (eNOS) mRNA expression, and myocardial oxidative and inflammatory markers. In vitro experiments were performed in isolated aortic and heart preparations. Eucalyptol improved angina manifestations, coronary flow, plasma NO, and aortic eNOS mRNA expression. It inhibited phenylephrine-induced contractions of endothelium-intact rings. Its relaxant effects decreased in L-NAME-pretreated rings and endothelium-denuded rings. Combination of eucalyptol and nifedipine exerted significant effects larger than each compound alone with blunting of nifedipine-induced tachycardia. The vascular eNO, antioxidant, and anti-inflammatory pathways were involved. This could lead to the development of a therapeutic antianginal system of both compounds.
Low-intensity exercise improves cardiac tolerance to ischemia/reperfusion injury in aged female rats with metabolic syndrome

Metabolic syndrome (MetS) is a group of cardiovascular risk factors which has higher prevalence in the elderly population. Being an aged obese woman with raised arterial blood pressure and blood glucose level might increase the risk of having cardiovascular morbidity or mortality during an attack of myocardial infarction. The present study was designed to evaluate the cardioprotective efficacy of a low intensity swim–exercise program on ischemic reperfusion injury in isolated perfused hearts from aged female rats with MetS. Wistar rats were allocated into three groups: controls, MetS–sedentary, and MetS–exercised. Body weight (BW) and waist circumference were measured with calculation of body mass index (BMI). Blood pressure, fasting blood glucose (FBG) and lipid profile were determined. Cardiac activities of isolated hearts were studied on Langendorff preparation under basal conditions and following ischemia/reperfusion (I/R), and the level of cardiac tissue nitrite was also assessed. MetS–exercised rats showed restoration of control values for BW, BMI, waist circumference, visceral fat, FBG, plasma lipid profile and left ventricular nitrite content. Meanwhile, arterial blood pressure was significantly lower than both controls and MetS–sedentary rats. These findings suggest that a short-term, low-intensity exercise program can improve cardiac tolerance to I/R injury possibly by ameliorating the high-risk cardiovascular components of the MetS.
Is the isolated heart a relaxation-oscillator?

The heart is the main organ of the circulatory system. It is in charge of carrying out two essential functions for life: providing nutrients to all cells and collecting carbon dioxide in the periphery of the body for its subsequent gaseous exchange. The isolated heart preparation allows a definitive evaluation of cardiac contractile function directly on the heart without interactions with other tissues and organs present in the body. This study aimed to investigate whether cardiac contraction can be considered as a relaxation-oscillator. When stimulating in the early stages of the contractile cycle, the results show a decrease in the time of occurrence of the poststimulus event, while in intermediate stages, the time of occurrence increases. However, the stimulus has a more significant impact on the temporality and morphology of the second postdisturbance event, resulting in an incomplete or barely noticeable contraction.
