Hog Cholera Virus (HCV) was maintained for over 400 days in cell cultures kept at optimum conditions by repeating the change of medium at intervals of 4–7 days and stripping off the aging parts of the cell sheet from the glass wall. No cytopathogenic effect of HCV was observed in this experiment.

Swine inoculated with cell-free fluid from the HCV-MIYAGI culture of 189–293 days developed no clinical reactions, nevertheless, they were able to withstand subsequent challenge with 10.000 MLD-ALD. The virulence of this cultured virus was remarkably reduced so as to cause no reactions in swine inoculated with 5.0 ml. of an undiluted culture, which was equivalent to 50.000 PU.

Immunity was produced with an injection of 1 ml. of 10−4 dilution already on the 7th day after inoculation. Despite the back passage of the attenuated virus over 10 times to susceptible adult swine no apparent increase in virulence was demonstrated.

The inoculated swine did not cause apparent contact infection among inmate susceptible swine.

Virulent strain ALD was attenuated just like low virulent strain MIYAGI and the attenuation steadily proceeded when it was treated by the bovine kidney continuous cell-virus propagation method.
