Immuntherapie der allergischen Rhinitis ohne Allergene?

Neue Möglichkeiten einer Immunmodulation durch Vakzinierung mittels „virus-like particles“ und CpG-Motiven

Zusammenfassung

Hintergrund

Die allergenspezifische Immuntherapie (SIT) wird allgemein als einzig kausaler Therapieansatz für die allergische Rhinitis akzeptiert. Bislang besteht als ein Dogma der Immuntherapie bei Inhalationsallergien, dass zumindest Allergenkomponenten für eine wirksame Therapie notwendig sind. Dieses Dogma könnte jedoch durch aktuelle Ergebnisse von effektiven Immuntherapien ohne Allergen ins Wanken kommen. „Virus-like particles“ (VLP) stellen einen neuen interessanten Aspekt für eine Immuntherapie bei Inhalationsallergien dar.

Ziel der Arbeit

Erste Erfahrungen zu erfolgreichen Immuntherapien ohne Allergenextrakte liegen vor. Im vorliegenden Beitrag werden die aktuell vorhandenen klinischen Erfahrungen mit Bakteriophagen-VLP, gefüllt mit CpG-Motiven, für die Behandlung der allergischen Rhinitis aufgezeigt.

Ergebnisse

VLP-Vakzine wurden als gut verträglich, immunogen und effektiv für die prophylaktische Impfung verschiedener Infektionen dargestellt. Bakteriophagen-VLP, gefüllt mit Oligonukleotiden, die CpG-Motive mit Tumorantigenen auf ihrer Oberfläche enthalten, führten klinisch zu einer Induktion von spezifischen T-Zellen bei Tumorpatienten und wurden erfolgreich als Adjuvanzien im Rahmen von prophylaktischen Vakzinierungen eingesetzt.

Schlussfolgerung

Die VLP-Technologie in Kombination mit der Anwendung von CpG-Motiven könnte dazu beitragen, komplexe Krankheiten wie maligne Erkrankungen, Autoimmunerkrankungen und Allergien ursächlich zu behandeln.
