Abstract

Subcellular fractionation studies were done using lactating rat mammary tissue. Lipoprotein lipase (E.C. 3.1.1.34) was found to be associated with plasma membrane-enriched and endoplasmic reticulum-enriched fractions. Considerable amounts of soluble lipoprotein lipase were found after homogenization of this tissue.

The membrane-associated lipoprotein lipase activities described here were found to be resistant to release, by heparin, from their membrane sites.

Buoyant density gradient fractionation experiments suggest that the lipoprotein lipase activity of lactating mammary tissue is located in part on the secretory epithelial cell plasma membrane. This conclusion is discussed in the context of the known site of action, in vivo, of mammary gland lipoprotein lipase, and of probable routes of its transport to that site.

There is a rapid stimulation of the rate of efflux of 3-O-methyl D-glucose from intact lactating rat mammary tissue slices in response to a change in the Na⁺-gradient across the basolateral plasma membrane. RNA extracted from lactating glands contains a 4 kb transcript which hybridises with the cDNA for rabbit intestinal SGLT1. A fraction of the mammary gland cells which is enriched in endoplasmic, Golgi and plasma membranes contains a protein that is immunologically similar to the Na⁺-dependent D-glucose symporter present in the plasma membrane of polarised epithelial cells, e.g., rabbit enterocytes, ovine enterocytes and parotid acinar cells.
Insulin-stimulated high affinity cyclic AMP phosphodiesterase in rat mammary acini

High affinity cyclic AMP phosphodiesterase activity in preparation of acini isolated from mammary tissue of lactating rats is shown to be stimulated by the addition of physiological concentrations of insulin to incubations of acini in vitro. This effect is expressed specifically on membrane-associated phosphodiesterase and occurs in the absence of concurrent protein synthesis. The possible functional role of this aspect of insulin's action on mammary tissue is discussed and compared with the well-known reversal by this hormone of the effects of lipolytic agents in adipose tissue and liver.
In vitro anti-sickling effect of cepharanthine

Cepharantine, a bisbenzylisoquinoline alkaloid, was found to have in vitro anti-sickling activity. The activity is of the same magnitude as that of chlorpromazine, an anti-psychotic drug, but cepharantine has no neuroleptic action. The most attractive feature of cepharanthine as a possible anti-sickling drug may be that the drug has been clinically used in Japan for other diseases and is known to have very few side effects both when given in large doses and over a long term. The drug can be administered either orally or intravenously. Cepharanthine has been reported to have several clinical effects, such as enhancement of the immune response, improvement of peripheral circulation, vasodilation and inhibition of platelet aggregation, all of which may also be beneficial to sickle-cell patients.
Placental lipoprotein lipase activity in the rabbit, rat and sheep

  1. The characteristics of lipolytic activity in various preparations derived from placentae of rabbits, rats and sheep have been compared with those of authentic lipoproteins lipases.

  2. A modification (shortening of incubation time) to standard lipoprotein lipase assays is required to measure placental alkaline lipase activity with accuracy.

  3. Placental triacylglycerol lipase activity is five-fold higher in the sheep than the rat and rabbit: all resemble lipoprotein lipase with respect to inhibition by NaCI and protamine sulphate, survival after acetone-ether treatment and pH-dependence.

  4. Placental lipoprotein lipase is atypical in its insensitivity to serum-stimulation.
