One mechanism of adenosine neuroprotection is via a reduction of microglia and macrophages activation (central and peripheral action), thereby reducing pro-inflammatory cytokines, chemokines and nitric oxide (Bischofberger et al., 1997; Cronstein, 1994) besides potently inhibiting TNF-α production (Le Vraux et al., 1993; Schubert et al., 1997). Likewise, a pro-inflammatory profile is well accepted as an important part of bipolar disorder pathophysiology (Jakobsson et al., 2015; Réus et al., 2015), including the presence of microglial activation (Stertz et al., 2013) and increased TNF-α production (Munkholm et al., 2013).

The ability of adenosine to modulate TNF-α production has been well known for a long time in human monocytes [17,18] and is consistent with the proclaimed general anti-inflammatory actions of extracellular adenosine. On the other hand, the present study demonstrates a dose-dependent stimulatory effect of adenosine on pro-inflammatory chemokine IL-8 production, which might imply that the effect of adenosine on monocytes is more multilayered and not strictly anti-inflammatory.
