Distribution of GABA-like immunoreactivity in the pigeon brain

Abstract

The distribution of GABA-like immunoreactivity in glutaraldehyde-fixed pigeon brains was studied by means of a monoclonal antibody. GABA-like immunoreactivity was observed in neuronal perikarya of different sizes as well as in neuropil and in certain fiber tracts. Certain staining patterns indicated the existence of several GABAergic projection systems in the pigeon brain. Indeed, a high density of immunostained perikarya and a low density of labeled terminal-like elements was the prominent pattern in the nuclei subpretectalis and posteroventralis, while an absence of perikaryal GABA-like immunoreactivity and accumulations of immunoreactive dots were observed in the isthmo-optic nucleus, amongst others. In the optic tectum, stained cell bodies with radially oriented processes in layer IIi (10) and with horizontally oriented processes in layer IId (5) were seen and were reminiscent of autoradiographic labeling patterns obtained previously following tectal injection of tritiated GABA. In the cerebellum, GABA-like immunoreactivity involved all types of neurons with the exception of granule cells. Purkinje cells showed regionally different intensities of immunostaining. In addition, in folium X no stained basket-like elements were observed.

Although there is no evidence as yet about the function of GABA in most of the structures, the present results indicate an important role for this neurotransmitter in the pigeon brain.

      The avian nucleus pretectalis (PT), a nonretinorecipient nucleus, receives a visual input from the optic tectum and has reciprocal connections with the SP/IPS, providing an inhibitory control of the rotundal activity (Tömböl et al., 1999; Theiss et al., 2003; Sun et al., 2005). The SP/IPS-PT complex is the main source of inhibitory GABAergic input to the Rot, thus playing a crucial role in the function of the tectofugal system (Benowitz and Karten, 1976; Hunt and Künzle, 1976; Domenici et al., 1988; Granda and Crossland, 1989; Bischof and Niemann, 1990; Tömböl et al., 1994, 1999; Mpodozis et al., 1996; Deng and Rogers, 1998; Hu et al., 2003; Theiss et al., 2003). The fundamental process of vision (the figure-ground discrimination) is associated with the activity of the Rot and SP/IPS (Acerbo et al., 2012; Scully et al., 2014).

      However, it is worth to note that these neurons are not only inhibitory interneurons, but also projection neurons belonging to the descending inhibitory cochleopetal system. In the mammalian (Fisher and Davies, 1976; Winer and Larue, 1996; Jones, 2007) and avian (Domenici et al., 1988; Müller, 1988; Veenman and Reiner, 1994; Sun et al., 2005; Pinaud and Mello, 2007) thalamic auditory center, a great diversity in number of interneurons (including their absence) with various morphological types have been observed. In the literature, data are extremely scarce with regard to interneurons in reptilian auditory centers.
