Brief noteThe vanB gene of vancomycin-resistant Enterococcus faecalis V583 is structurally related to genes encoding d-Ala:d-Ala ligases and glycopeptide-resistance proteins VanA and VanC☆

Mobile genetic elements beyond the VanB-resistance dissemination among hospital-associated enterococci and other Gram-positive bacteria
2021, PlasmidCitation Excerpt :This phenotype was associated with an acquisition of the vanA gene cluster, located on Tn1546-type transposons (Arthur et al., 1993). The following years brought identification of other classes of van genes, including vanB in the V583 strain of E. faecalis (Sahm et al., 1989; Evers et al., 1993) and in the D366 isolate of E. faecium (Williamson et al., 1989), as well as vanC, vanD, vanE, vanG, vanL, vanM and vanN (reviewed recently by Gorrie et al., 2019). Of these nine determinants, vanA and vanB clusters are of the largest epidemiological significance, leading over time to the growing levels of vancomycin-resistant enterococci (VRE) reported worldwide.
From Natural Products to Drugs: Glyco- and Lipoglycopeptides, a New Generation of Potent Cell Wall Biosynthesis Inhibitors
2016, Studies in Natural Products ChemistryCitation Excerpt :VanY encodes a membrane-associated carboxypeptidase that hydrolyzes the peptidoglycan peptide side chain to which vancomycin usually binds [42]. VanS and VanR are involved in regulation of VanA, VanH, VanX, and VanY gene expression [43]. Vancomycin has very low oral bioavailability and is used orally to treat C. difficile associated diarrhea (CDAD) administering 125–500 mg every 6 h either using the injectable solution or a capsule.
