The solute carrier family 11 member A1 (SLC11A1) gene, formerly known as natural resistance-associated macrophage protein 1 (NRAMP1) (Vidal et al., 1996), encodes an integral membrane protein regulating the activity of macrophages (Blackwell, 1996; Forbes and Gros, 2001). The SLC11A1 protein has 12 transmembrane domains (TM1–TM12), several phosphorylation sites and three functional domains, namely a Src (sarcoma) homology 3-binding motif (SH3), a glycosylation site and a transport motif (Feng et al., 1996).

SLC11A1 exerts pleiotropic effects on macrophage function, including increased expression of inducible nitric oxide synthase (iNOS) and subsequent generation of nitric oxide (NO), upregulation of MHC class II expression and enhanced antigen presentation to T cells, increased production of pro-inflammatory cytokines (notably interleukin [IL]-1β and tumour necrosis factor [TNF]-α), production of reactive species involved in oxidative burst, and upregulation of KC (a C-X-C chemokine, belonging to the IL-8 family, that is chemotactic for neutrophils) [26–32]. Expression of Slc11a1 in the late endosomal/lysosomal compartments of murine dendritic cells has been recently reported [33].
