Marginale Protektion retinaler Zellen durch Bisperoxovanadium

Geeignete Therapie im Modell der retinalen Ischämie?

Zusammenfassung

Hintergrund

Ischämische Prozesse führen in der Regel zu einer Destruktion retinaler Zellen und spielen bei vielen Augenerkrankungen eine entscheidende Rolle.

Ziel der Arbeit

Bisperoxovanadium sollte erstmals hinsichtlich eines potenziellen neuroprotektiven Effektes in einem Tiermodell der retinalen Ischämie untersucht werden.

Material und Methoden

Initial wurde in einem Auge eine Ischämie mithilfe eines Ischämie/Reperfusionsmodells induziert. Drei Tage später folgte eine 14-tägige medikamentöse Behandlung bei einem Teil der Tiere. Dafür wurde Bisperoxovanadium alle 3 Tage intraperitoneal appliziert. Im Anschluss erfolgte die Auswertung der Ganglienzellzahl, der Apoptoserate, der Amakrinzellen, der Makroglia, der Mikroglia und ihres Aktivierungszustandes sowie der Photorezeptoren mittels Histologie und Immunhistochemie.

Ergebnisse

Bei den ischämischen Augen konnten im Vergleich zur Kontrollgruppe ein signifikanter Ganglienzellverlust, eine signifikante Reduktion der inneren Schichten sowie eine Abnahme der Photorezeptorenzahl und der Amakrinzellen ermittelt werden. Gleichzeitig kam es zu einer Zunahme der Anzahl an Mikroglia. Bei den mit Bisperoxovanadium behandelten Tieren stellte sich kein signifikanter neuroprotektiver Effekt in Bezug auf Ganglienzellzahl, Apoptoserate, Makroglia, Amakrinzellen und Photorezeptoren ein. Es konnte jedoch eine geringere strukturelle Degeneration der Photorezeptoren beobachtet werden. Auch bei den Mikroglia und ihrer aktivierten Form wurden niedrigere Zahlen nach Behandlung mit Bisperoxovanadium festgestellt.

Diskussion

Bisperoxovanadium scheint sich nur marginal neuroprotektiv auf die ischämische Retina auszuwirken. Es wäre zu prüfen, ob früherer Therapiebeginn, höhere Dosis oder eine andere Verabreichungsform diese Effekte verbessern können oder ob dieser Therapieansatz nicht geeignet ist.
