Loss of histone H3 trimethylation (H3K27me3) is a hallmark feature of histone H3 K27M-mutated diffuse midline gliomas, but reduced H3K27me3 may also be encountered in tumor entities only rarely showing K27M mutations. Panwalkar et al. recently demonstrated an important diagnostic and prognostic role of reduced H3K27me3 expression in a molecular subgroup of ependymoma, i.e., group-A childhood posterior fossa ependymoma [6]. …
