Welche Mechanismen führen zur Triploidie?

Erkenntnisse aus der zytogenetischen Analyse in vitro entstandener, abnormer Vorkernstadien

Zusammenfassung

Triploidie bedeutet das Vorliegen von 3 anstelle von 2 haploiden Chromosomensätzen in einer Zelle. Basierend auf Erkenntnissen aus eigenen zytogenetischen Untersuchungen an abnorm befruchteten Eizellen werden wichtige Mechanismen vorgestellt, die in vitro zur Bildung triploider Zygoten führen. Die Kombination von 2 paternalen Anteilen mit einem maternalen Chromosomensatz (Diandrie) kann durch das Eindringen von 2 individuellen, haploiden Spermatozoen in eine Eizelle zustande kommen oder durch die Befruchtung einer Eizelle mit einem diploiden Spermatozoon. Während diese Anomalie im ersten Fall infolge Ausbildung von 3 Vorkernen offensichtlich ist, wird beim 2. Mechanismus die reguläre Zahl von 2 Vorkernen zu beobachten sein, weshalb eine Erkennung der betroffenen Zelle anhand dieses Kriteriums nicht gelingt. Die Kombination von 2 maternalen Anteilen mit einem paternalen Chromosomensatz (Digynie) kann auf einer unterbliebenen Ausstoßung des 2. Polkörpers beruhen. Eine derartige Aberration tritt gelegentlich nach intrazytoplasmatischer Spermieninjektion (ICSI) auf und gibt sich durch 3 Vorkerne zu erkennen. Weniger geläufig ist hingegen die durch diploide Rieseneizellen verursachte digyne Triploidie. Hier können, abhängig vom Verlauf der Reifung zur Metaphase II, nach dem Eindringen eines Spermatozoons entweder 3 oder nur 2 Vorkerne entstehen. Im letzten Fall ist die Anomalie dann zu erkennen, wenn neben der Vorkernzahl die Größe der Zelle im Vorkernstadium als zusätzliches Beurteilungskriterium herangezogen wird.
