Die Rolle von Tyrosinkinasen bei Krebserkrankungen des Kopf-Hals-Bereichs

Zusammenfassung

Rezeptortyrosinkinasen spielen eine zentrale Rolle in der interzellulären Kommunikation zum Erhalt der Integrität der Zellen im Zellverband. Genetische Veränderungen in diesen Rezeptor-Enzym-Komplexen bzw. in den sich anschließenden Signalkaskaden können zu einer Enthemmung charakteristischer zellulärer Eigenschaften wie Proliferation, Adhäsion, Differenzierung und Apoptose führen. Als Folge erklärt sich die Rolle dieser Rezeptorenklasse in der Pathogenese maligner Erkrankungen. Der Artikel resümiert die molekularen Grundlagen und Gruppeneinteilungen dieser Rezeptorenfamilie und setzt sich nachhaltig mit der Literaturaufarbeitung über die Rolle der Rezeptoren in der Karzinogenese auseinander. Als Exempel werden die Aspekte der Fibroblastenwachstumsfaktorrezeptoren (FGFR) diskutiert, ebenso der Stellenwert von sog. Einzelnukleotidpolymorphismen in der Krebsentstehung. Die Autoren zeigen, dass der Ansatz einer molekularen Therapie vielversprechend ist und im Rahmen einer multimodalen Krebstherapie in Betracht gezogen werden sollte.
