Orchestrierung der Entzündung bei Kontaktallergie durch angeborene Immun- und zelluläre Stressantworten

Zusammenfassung

Hintergrund: Entzündung ist von zentraler Bedeutung für die Initiation von Immunantworten und für die Pathogenese zahlreicher Erkrankungen wie der allergischen Kontaktdermatitis („allergic contact dermatitis“, ACD). Die ACD ist eine entzündliche Hauterkrankung, die von niedermolekularen organischen Chemikalien und Metallionen verursacht wird. Dabei spielt das Immunsystem die entscheidende Rolle. Die auslösenden Chemikalien wirken nach Proteinbindung als Kontaktallergene, die von spezifischen T-Zellen erkannt werden. Zuvor müssen die Chemikalien aber eine Entzündung in der Haut auslösen, ohne die vor allem das adaptive Immunsystem nicht aktiviert wird.
Methoden: In den letzten Jahren wurden die Entzündungsmechanismen der Kontaktallergie auf zellulärer und molekularer Ebene in vivo und in vitro untersucht.
Ergebnisse: Kontaktallergene aktivieren das angeborene Immunsystem und zusätzlich zelluläre Stressantworten, die im Zusammenspiel für die Hautentzündung verantwortlich sind. Dabei wird die Entzündung sowohl für die initiale Sensibilisierung als auch für die Auslösung der allergischen Kontaktdermatitis benötigt.
Diskussion: Die Hautentzündung bei der allergischen Kontaktdermatitis wird durch das Zusammenspiel von angeborenem Immunsystem und zellulären Stressantworten orchestriert.
