Der Beginn der modernen Opioidforschung liegt im Jahr 1804, als der deutsche Chemiker und Apotheker Sertürner das Alkaloid Morphin aus dem Opiumsaft isolieren konnte. Über Fentanyl und den modifizierten Fentanylderivaten bis hin zu den neuesten Entwicklungen konnten bis heute zahlreiche analgetisch potente Substanzen synthetisch hergestellt werden, deren Pharmakologie und klinische Anwendung in diesem Kapitel erörtert werden.
