Tegafur is an oral fluoropyrimidine. When given continuously for several weeks, in combination with uracil, plasma concentrations of fluorouracil are reached similar to those obtained with continuous intravenous infusion of fluorouracil. Two randomised studies demonstrated that the response rate and survival were similar with tegafur/ uracil plus calcium folinate and fluorouracil plus folinic acid in patients with advanced colorectal cancer. Ongoing studies will determine whether the outcome of these patients may be improved by adding oxaliplatin, irinotecan or other cytostatic agents. Tegafur/uracil is also active in tumours of the head and neck, stomach and breast. …
