Association between beta defensin gene polymorphism and clinical endometritis in dairy cows

Abstract

Defensins especially β-defensins participate in innate immunity to prevent microbial infection of female reproductive system, one of the most important problems in mammal species. So, the purpose of this study was to investigate the presence of polymorphism in β-defensin gene (C/T: position 2239) of Iranian dairy cattle (n = 127) and evaluate its effect on susceptibility to clinical endometritis. To recognize this point mutation, the RFLP-PCR method was used. As a result, three distinct genotypes of cattle (CC, CT, and TT) were determined based on this locus. In this study, the genotype frequencies of this SNP were 59.1%, 32.3%, and 8.7% for CC, CT, and TT, respectively. Also, allele frequencies were 75.2% and 24.8% for C and T, respectively. Results indicate that cows having Allele T had desirable resistance against clinical endometritis based on their history fertility in which the clinical endometritis occurred with significantly less frequency (p = 0.008) in cows having Allele T (61.5%) compared to those lacking Allele T (82.7%). In conclusion, our study suggested that the C to T transition within the intronic region of β4-defensin gene (position 2239) might be useful for increasing the resistance of reproductive system in Holstein dairy cows.
