Neue Therapieansätze für die Multiple Sklerose?

Zusammenfassung

Einer der kritischen Schritte in der pathogenetischen Kaskade der Multiplen Sklerose (MS) ist die Einwanderung von mononukleären Zellen (Lymphozyten und Monozyten) in das zentrale Nervensystem (ZNS). Aufgrund ihrer Beteiligung an der Migration von Leukozyten unter physiologischen und pathologischen Bedingungen liegt eine Beteiligung von Chemokinen an diesem Prozess nahe. Dieses komplexe System von Liganden und Rezeptoren scheint für ein therapeutisches Eingreifen besonders interessant, da neben Peptidantagonisten auch kleine molekulare Antagonisten für den klinischen Gebrauch zur Verfügung stehen. Zu den möglicherweise bei der MS therapeutisch interessanten Chemokinrezeptoren gehören CCR1, CCR2, CCR5 und CXCR3 und ihre entsprechenden Liganden.
Dieser Übersichtsartikel fasst den heutigen Wissensstand zu der Rolle von Chemokinen und Chemokinrezeptoren in der Pathogenese der MS zusammen und eröffnet erste Ansätze der auf diesen Kenntnissen beruhenden pharmakologisch-therapeutischen Interventionsmöglichkeiten, die in Kürze auch in klinischen Studien an MS-Patienten geprüft werden.
