Zusammenfassung

Ein Therapieansprechen, das mit Computertomographie (CT) oder Magnetresonanztomographie (MRT) gemessen wird, ist ein früher Parameter für die Effizienz einer Tumortherapie, korreliert jedoch nicht in jedem Fall mit dem Gesamtüberleben. Mögliche Erklärungen dafür sind die Entstehung von Resistenzen während der Therapie und der Einsatz verschiedener Substanzen über mehrere Therapielinien im Behandlungsverlauf. Eine funktionelle Bildgebung mit der Kombination aus Positronenemissionstomographie (PET) und CT ist eine vielversprechende Methode zur frühen Bewertung des Ansprechens und der Vorhersage von Überlebensendpunkten. Basierend auf einem tieferen Verständnis der Tumorbiologie stellen die Prädiktoren der Effizienz bestimmter zielgerichteter Therapien auch molekulare Marker für das Ansprechen dar. Darüber hinaus wurden Modelle zur Vorhersage der Tumor-Response entwickelt, die über reine Zellkulturtechniken hinausgehen (Tumorschnittkulturen, Xenograft-Modelle). Diese werden aktuell intensiv weiterentwickelt. Genexpressionsanalysen an Tumorgewebe werden zur Identifizierung von prädiktiven Genexpressionsprofilen ebenfalls verwendet. Hier steht eine weitere klinische Validierung noch aus.

Finale OS-Analyse der MONARCH-3-Studie vorgestellt

In der MONARCH-3-Studie erhielten Patientinnen mit fortgeschrittenem HR+, HER2- Brustkrebs Abemaciclib [1,a] in Kombination mit nicht-steroidalem Aromatasehemmer (nsAI). Die finalen Daten bestätigen den in früheren Analysen beobachteten Unterschied zugunsten der Kombinationstherapie. [2] Details dazu vom SABCS 2023.
