Glycine transporter of canine lens epithelial cells: functional analysis and molecular structure

Abstract

Glycine transport activity in a canine lens epithelial cell line was investigated. The glycine transport activity was 2.70 ± 0.257 nmol/mg protein/min at 100 μM extracellular glycine concentration, which was reduced to 9 % in a sodium-free condition. ALX5407 (analogue of sarcosine), the specific inhibitor of the glycine transporter 1 (GlyT1), reduced 30 % of the transport activity at 100 nM; on the other hand, ALX1393, the glycine transporter 2 (GlyT2) inhibitor, did not affect glycine transport activity of lens epithelial cells. A full-length cDNA sequence of canine GlyT1 in lens epithelial cells was 2218 bp long and encoded 620 amino acid polypeptides. The deduced amino acid sequence of canine GlyT1 indicated high homology to bovine E isotype of GlyT1.
