Einsatz von 2-D- und 3-D-Kulturen in der Dermatologie

Zusammenfassung

Die Haut ist ein komplexes Organ, das eine Reihe lebenswichtiger Funktionen erfüllt, indem es u. a. eine physikalische Barriere bildet, die unseren Körper vor dem Eindringen von Pathogenen und Irritanzien sowie vor übermäßigem Flüssigkeitsverlust schützt. Neben den passiven Eigenschaften ist die Haut auch aktiv am Immungeschehen beteiligt. Ein komplexer Aufbau aus unterschiedlichen Zelltypen und Strukturen erlaubt es der Haut, diese Funktionen zu erfüllen. Die In-vitro-Forschung steht oft vor dem Problem, dass einfache 2‑D-Zellkulturen diese Funktionen nicht ausreichend abbilden können. 3‑D-Hautmodelle bieten hier Abhilfe. In den letzten Jahren kam es zu einer bedeutsamen Weiterentwicklung auf diesem Gebiet; die Reproduzierbarkeit der Methode sowie die physiologische Struktur und Gewebearchitektur der 3‑D-Hautmodelle wurden verbessert. Je nach Fragestellung sind heute Protokolle für 3‑D-Hautmodelle publiziert, die vom einfachen mehrschichtigen Epidermismodell bis zum höchst komplexen vaskularisierten 3‑D-Vollhautmodell reichen.
