Epigenetische Mechanismen der Depression

Zusammenfassung

Der Begriff Epigenetik beschreibt Mechanismen, welche die Funktion von Genen verändern können, ohne eine zugrunde liegende Veränderung der eigentlichen DNA-Sequenz. Zu den epigenetischen Mechanismen gehören u. a. die Histonproteinmodifizierung (Methylierung, Acetylierung, Phosphorylierung) und die DNA-Methylierung. Histonmethylierung und -deacetylierung an Promoterregionen von wichtigen mit der Depression in Zusammenhang stehenden neurotrophen Faktoren führen zu deren verringerten Expression. Die DNA-Methylierung in Promoterregionen von Rezeptor- und Neurotrophingenen führt ebenfalls zu deren verminderter Expression, wie für die Depression gezeigt werden konnte. Tierexperimentelle Studien haben gezeigt, dass maternales Aufzuchtverhalten über epigenetische Mechanismen die Stressreaktivität der Hypothalamus-Hypophysen-Nebennierenrinden-Achse der Nachkommen entscheidend beeinflusst. Dabei handelt es sich um erworbene Veränderungen, die unter medikamentöser (antidepressiver) Behandlung teilweise rückgängig gemacht werden können.
