Zusammenfassung

Epigenetische Mechanismen wie DNA-Methylierung oder Histonmodifikationen sind biochemische Veränderungen an der DNA oder ihrer Raumstruktur. Sie steuern die Genfunktion, werden durch Umweltfaktoren beeinflusst und sind zeitlich dynamisch. In dieser Übersichtsarbeit wird der aktuelle Forschungsstand zur Rolle der Epigenetik in der Pathogenese psychischer Erkrankungen exemplarisch für Schizophrenie, Depression, Angsterkrankungen und posttraumatische Belastungsstörung zusammengefasst. Weiterhin werden epigenetische Modifikationen unter einer pharmako- und psychotherapeutischen Behandlung dargestellt. Epigenetische Mechanismen nehmen eine zentrale Scharnierfunktion an der Schnittstelle zwischen Genen und Umwelt und damit im Vulnerabilitäts-Stress-Modell psychischer Erkrankungen ein. Zukünftig könnten epigenetische Profile im Sinne eines „precision medicine“-Ansatzes als Marker des Erkrankungsrisikos und des Therapieansprechens nutzbar werden und mögliche „druggable targets“ darstellen.
