TNFα-Inhibitoren in der Uveitistherapie

Zusammenfassung

Der Tumor-Nekrose-Faktor α (TNFα) ist ein pleiotropes Zytokin, das eine Schlüsselrolle in der Pathophysiologie chronisch-entzündlicher Erkrankungen spielt. Tierexperimentelle Untersuchungen zeigen, dass TNFα auch bei der nichtinfektiösen Uveitis eine wichtige Komponente proinflammatorischer Prozesse ist. Daher ist die gezielte Blockade von TNFα durch TNF-Rezeptorfusionsproteine oder monoklonale Antikörper ein vielversprechender Ansatz in der Uveitistherapie. Zudem haben die aktuell verfügbaren TNFα-Inhibitoren im Vergleich zu den konventionellen Immunsuppressiva ein günstiges Nebenwirkungsprofil. Die bisherigen Studien zeigen eine gute Effizienz von TNFα-Inhibitoren in der Therapie der posterioren Uveitis. Die aktuelle Datenlage lässt jedoch noch keine endgültige Aussage über die Wirksamkeit der TNFα-Inhibitoren bei der anterioren Uveitis zu.
