Genetisch bedingte Haarerkrankungen

Zusammenfassung

Patienten mit Haarverlust und unerwünschtem übermäßigem Haarwachstum sind eine große Herausforderung für den Dermatologen, da die Ätiopathogenese der meisten Haarerkrankungen nicht hinreichend bekannt ist und die therapeutischen Möglichkeiten limitiert sind. Dies gilt insbesondere für genetisch bedingte Haarerkrankungen, bei denen alle gegenwärtigen Behandlungsversuche frustran sind und die aufgrund der ausgeprägten klinischen und genetischen Heterogenität auch eine diagnostische Herausforderung darstellen. Die enormen Fortschritte in der molekularbiologischen Forschung während der zurückliegenden 20 Jahre, insbesondere die verschiedenen neuen Techniken auf dem Gebiet der Hochdurchsatzsequenzierung kompletter Exome und Genome, haben uns jedoch in die Lage versetzt, die genetische Basis der meisten monogen vererbten Haarerkrankungen zu entschlüsseln, wenn hierfür ein oder mehrere betroffene Indexpatienten bzw. Familien sowie die entsprechenden apparativen und finanziellen Mittel zur Verfügung stehen. In dieser Übersicht geben wir ein Update zu Klinik und Genetik ausgewählter monogen und polygen vererbter Erkrankungen, die sich mit Hyper- oder Hypotrichose manifestieren.
